Phenylephrine Hydrochloride BP 5 mg
Chlorpheniramine Maleate BP 2 mg
Flavoured syrupy base q.s.
DESCRIPTION: KOFF-GO MINT is a cough reliever formula containing Dextromethorphan for dry unproductive cough or if cough is unduely tiring or hazardous, Phenylephrine Hydrochloride used as decongestant and Chlorpheniramine Maleate an alkylamine derivative is an antihistamine that causes a moderate degree of sedation.
MECHANISM OF ACTION: They afford relief in cough due to their action in the CNS to raise the threshold of cough centre or act peripherally in the respiratory tract to reduce tussal impulses, Chlorpheniramine have sedative & anti cholinergic actions, but lack selectivity for the cough centre. Chlorpheniramine blocks the effects of the naturally occurring chemical histamine in your body. Chlorpheniramine is used to treat sneezing; runny nose; itching, watery eyes; hives; rashes; itching; and other symptoms of allergies and the common cold. Phenylephrine is an al -adrenergic receptor agonist used primarily as a decongestant.
PHARMACOKINETICS: Dextromethorphan & Phenylephrine are readily & completely absorbed from G.I. tract, peak plasma concentrations can be attained in 1-2 hours & both are excreted unchanged in urine. Dextromethorphan is well absorbed from the gastrointestinal tract with maximum serum level occurring at 2.5 hours. Peak concentration of the major metabolite dextrorphan was 1.6 to 1.7 hours. Onset of effect is rapid, often beginning 15 to 30 minutes after oral ingestion. Chlorpheniramine Maleate absorbed slowly from G.I. tract, peak plasma concentrations about 2.5 to 6 hours after administration. Bioavailability is low, 25 to 50 % reported. Widely distributed in the body including CNS.
INDICATIONS: Complete relief from dry cough & cough due to cold. Nonproductive cough caused by a cold and flu, or other conditions, decongestant, Sneezing; runny nose; itching, watery eyes; hives; rashes; and other symptoms of allergies and the common cold.
DOSAGE AND ADMINISTRATION:
Children over 2 years: 1 teaspoonful (5 ml) every 4 hours.
Adults: 2 teaspoonfuls (10 ml) every four hours.
CONTRAINDICATIONS: Hypersensitivity to any ingredients of the preparation.
PRECAUTIONS: Shake well before use. Avoid over medication. May cause drowsiness in an occasional patient, so sedative materials like alcohol should be avoided. Keep all medicines out of reach of children
DRUG INTERACTIONS:
Hypertensive crisis in patients receiving MAOIs. & drugs like amiodarone, fluoxetine, haloperidol affects the metabolism. Enhance the effects of sedatives like alcohol, barbiturates, hypnotics, opioid analgesics.
SIDE-EFFECTS: Dizziness, dry mouth, G.I.disturbances. Excitation, confusion & respiratory depression may occur after overdose.
STORAGE: Store at a temperature not exceeding 30°C. Protect from light.
