MECHANISM OF ACTION:
Domperidone facilitates gastrointestinal smooth muscle activity by inhibiting dopamine at the D1 receptors and inhibiting the release of neural acetylcholine by blocking D2 receptors. It decreases short bowel transit time by increasing oesophagal and gastric peristalsis and lowering oesophagal sphincter pressure. The antiemetic properties are related to its dopamine receptor-blocking activity at both the chemoreceptor trigger zone and the gastric level. It has strong affinities for the D2 and D3 dopamine receptors, which are found in the chemoreceptor trigger zone, located just outside the blood-brain barrier, which regulates nausea and vomiting.
INDICATIONS:
Treatment of nausea and vomiting of various origins, including infectious, dietetic origin, or induced by radiotherapy and slowed movement in the gastrointestinal tract.
CONTRAINDICATIONS:
In patients with moderate or severe hepatic impairment, known hypersensitivity to domperidone or any of the excipients.
SIDE EFFECTS:
Headache, dizziness, dry mouth, nervousness, flushing, or irritability may occur during the first several days as your body adjusts to the medication.
PRECAUTIONS:
Shake well before use. Avoid overmedication.
